TC Acclimating
Tissue Culture Acclimation Schedule
(Fresh Out of Flask)

Rinse: Carefully wash off agar with lukewarm distilled or clean water.

Medium: Plant in a fine, airy, sterile mix (sphagnum moss, perlite, or aroid mix depending on species).

Humidity: 95–100% (use a sealed clear container, plastic dome, or zip-lock bag).

Light: Bright indirect light, 10–12 hours (avoid direct sun).

Temperature: 75–80°F (24–27°C).

Water: Keep medium moist, not soggy.
Week 1

Start venting the container: open lid/zipper for 10–15 minutes once or twice a day.

Humidity stays high (90–95%).
-
Watch for leaf wilt — if severe, reduce venting.
Week 2

Increase venting gradually: 30–60 minutes twice daily.

Humidity ~85–90%.

Plants start thickening leaves, roots begin adapting.
-
Maintain bright indirect light.
Week 3

Leave lids cracked or partially open most of the day.
-
Humidity ~75–85%.
-
By now plants should show some growth (new leaves, root tips extending).
Week 4

Move to open air in grow tent or greenhouse conditions.
-
Humidity ~70–75% (good for most tropicals).

Water normally, allow medium surface to dry slightly between waterings.

Begin light fertilization (¼ strength).
After Week 4

Plants are usually hardened off enough to handle “normal” aroid or houseplant care.

Gradually adapt them to your final environment (room humidity or greenhouse).

Avoid direct sun until they are stronger.
⚠️ Tips & Warnings:

Some species (like anthuriums, alocasias, philodendrons) may need longer hardening.

If you see collapse/wilting, increase humidity again and slow down.

Don’t fertilize too early — roots are sensitive at first.
